Introduction of Protein A Affinity Filler
Protein A affinity chromatography packing is widely used for affinity purification of monoclonal antibodies or Fc fusion proteins because of its high selectivity. Due to the different elution performance, the affinity purification process of Protein A needs more development work, and has a close relationship with downstream process development. JSR Life Science has carried out a series of research and development work on this affinity chromatography filler, and introduced Amsphere TM A3, a very high quality filler.
Protein A has the ability to bind to antibodies without binding sites to antigens. Protein A can be specifically bound to the Fc segment of the antibody IgG molecule, and Protein A can be coupled to agarose gel as a ligand. When the samples such as antiserum and cell supernatant flow through it, the specific Fc segment of IgG will be bound to the ligand, and other proteins will flow through and after cleaning. The required IgG with high purity can be recovered by elution with corresponding buffers.
However, when the purified antibody is different, the binding capacity of different host antibodies is different. We also need to treat it according to the specific circumstances. We recommend the mAb IgG2a, IgG2b, IgG3, rabbit, human and pig polyclonal antibody to be purified by Protein A. Protein A affinity chromatography has become a widely used method for antibody purification. It can isolate and purify various mammalian antibodies of different subtypes or recombinant proteins containing Fc fragments from ascites, serum and cell culture supernatants or cell extracts.
